Epithelial cells, when fully polarized on porous support, exhibit many activities that require lots of energy input.

By providing ample energy source, epithelial cells will:

(1) Form monolayer with little lateral spaces

(2) Generate apical, lateral, and basal plasma domains with distinct protein composition

(3) Develop specific cell-cell interactions including zonula occludens, macula occluden, zonula adherens, macula adherens

(4) Polarize cytoplasmic contents including basal nucleus, golgi, actin and microtubular networks, terminal web

(5) Provide selective permeability barrier to ions and macromolecules through the tight junction

(6) Perform vectorial transport of ions, secretion of growth factors, transcytosis of receptor-mediated and fluid phase molecules

FEED CELLS GENEROUSLY:

- Tissue Culture Dish: ~25 mL media per 100 mm confluent cells every two days
- Transwells 12mm: ~ 3mL (2mL basal + 1 mL apical) per confluent cells everyday
